⑷ Basic requirements of the procurement :
A High acceleration voltage cryo- electron microscope
a The electron gun is a cold cathode field emission type electron gun with an acceleration voltage of 200kV.
b The sample introduction method is a cartridge introduction method, and it has a function to automatically transport the frozen sample into the column.
c Sample loading and unloading can be performed while maintaining the vacuum state of the sample transfer chamber by equipping the sample transfer mechanism with an airlock valve.
d Must have an in-column energy filter.
e The amount of electron beam dose can be continuously modulated by a high-speed electrostatic shutter.
f Equipped with an direct electron detection type camera, the number of effective pixels of the detector is 4,000×4,000 pixels or more, and the rate is 1500 images/second or more in full .
B Automatic rapid freezing device
a The temperature of the environmental chamber can be adjusted in the range of +4℃ to +60℃.
b Must have a mechanism to humidify the environmental chamber.
c The operation must be a touch screen or a foot switch.
d The temperature of the chamber that holds the refrigerant can be adjusted in the range of -196℃ to 0℃.
e The liquid nitrogen storage container must have a bakeout mechanism.
C Glow discharge surface modifier
a Provide a chamber that uses the atmosphere and a chamber that uses the vapor of the compound.
b Safe vapor supply of compounds.
c Must have a mechanism to prevent cross contamination between chambers.
